Oriscience Annexin V-FITC/PI Apoptosis Detection Kit provides a fast and convenient method for detecting apoptosis, which can be detected by flow cytometry or fluorescence microscopy. Annexin V is a calcium-dependent phospholipid binding protein that has a high affinity for phosphatidylserine (PS). Under normal physiological conditions, PS is mainly located in the inner leaflet of the plasma membrane. When apoptosis is initiated, PS is transferred to the outer cell membrane. Once PS reaches the outside of the membrane, it can be bound by Annexin V-FITC, thereby detecting cell apoptosis. Propidium iodide (PI) can specifically bind to double-stranded DNA and produce strong fluorescence, and cannot normally penetrate the cell membrane. Due to the loss of integrity of the cell membrane in the late stage of apoptosis or necrotic cells, PI can enter the cell to stain DNA. When used in combination with Annexin V, cells in different stages of apoptosis can be distinguished.
