Oriscience Protease Phosphatase Inhibitor Cocktail, 100× is a universal protease and phosphatase inhibitor cocktail. The core components of this product are a tube of protease inhibitor cocktail (tube A, 100× in DMSO) and a tube of phosphatase inhibitor cocktail (tube B, 100× in ddH2O). Tube A consists of 6 protease inhibitors, AEBSF, Aprotinin, Bestatin, E-64, Leupeptin, and Pepstatin A, which have a broad spectrum of protease inhibition. Tube B consists of 5 phosphatase inhibitors, Sodium Fluoride, Sodium Orthovanadate, Sodium Tartrate Dihydrate, Sodium Molybdate, and Imidazole, which can broadly inhibit the activity of acid phosphatases, alkaline phosphatases, and protein tyrosine phosphatases (PTPs) and are used to maintain the phosphorylation state of proteins. Separately packaged EDTA is also provided for selective use. If it is used to detect the activity of metalloproteinases, it is not advisable to add EDTA.
This product can effectively inhibit various proteases (such as serine proteases, aminopeptidases, cysteine proteases, threonine and aspartic acid proteases, metalloproteinases, etc.) and phosphatases (such as serine/threonine, tyrosine, acid and alkaline phosphatases, etc.) in conventional cell or tissue extracts. AEBSF is a water-soluble irreversible inhibitor of serine proteases, and its inhibition constant is similar to that of PMSF and DFP. It can effectively inhibit proteases such as trypsin, chymotrypsin, plasmin, kallikrein and thrombin. As a substitute for PMSF and DFP, AEBSF has lower toxicity, better water solubility and aqueous solution stability. Aprotinin is a competitive reversible inhibitor of serine proteases, Bestatin is a reversible inhibitor of aminopeptidase, E64 is an irreversible inhibitor of cysteine proteases, Leupeptin is a reversible inhibitor of serine and cysteine proteases, Pepstatin A is a reversible inhibitor of aspartic proteases, EDTA as a chelating agent is a reversible inhibitor of metalloproteinases, sodium fluoride is a reversible inhibitor of acid phosphatase, sodium orthovanadate is a reversible inhibitor of alkaline phosphatase and tyrosine phosphatase, sodium tartrate dihydrate is a reversible inhibitor of acid phosphatase, sodium molybdate is an irreversible inhibitor of acid phosphatase, imidazole is a reversible inhibitor of alkaline phosphatase.
This product can be widely used in protein immunoassay (WB), immunoprecipitation (IP), co-immunoprecipitation (Co-IP), pull-down, immunohistochemistry (IHC), immunofluorescence (IF), kinase research and other analyses.
