This product is a 5×SDS-PAGE protein loading buffer, a modified 5x concentrated protein loading buffer using bromophenol blue as an indicator dye, suitable for preparing protein samples for routine SDS-PAGE gel electrophoresis. Its mechanism of action is as follows: SDS in the buffer binds to proteins, forming a protein-SDS complex with a large number of negative charges, thus masking the protein's own charge differences and making its migration rate in an electric field dependent only on its molecular weight. Simultaneously, SDS breaks the hydrogen bonds of proteins, causing their secondary and tertiary structures to dissociate, while the reducing agent DTT breaks the disulfide bonds between cysteine residues, completely disrupting the protein's spatial conformation. Bromophenol blue, as an indicator during electrophoresis, can be used to monitor and predict the electrophoresis endpoint in real time.
